چکیده

Background: Trinucleotide repeats CAG (n) in androgen receptor gene is thought to be central to PCOS genetic susceptibility. However, previous studies of PCOS have shown variable association of CAG (n) polymorphism with PCOS. Objective: To assess the association of the AR gene CAG repeat length polymorphism with PCOS among Egyption patients, and its influence on clinical and biochemical androgen traits. Patients and Method: A cohort of 80 women with oligoanovulatory cycles and ultrasound features of PCOS and 67 healthy women of reproductive age were investigated for CAG repeat lengths using genescan. Assay of serum total testosterone, free testosterone and DHEAS were carried out. These parameters were correlated with the CAG repeat. Results: AR (CAG) n range were 8-25 with a median of 19 in PCOS patients and 10-29 with a median of 22 in control group. The difference in median repeat number was significant comparing PCOS with control ( p<0.001). A higher frequency of short AR (CAG) n alleles ( 9 15) tended to be more frequent in PCOS women than controls (p=0.005). We found significant difference between the both groups as regard BMI, Hirsutism, Total testosterone, Free testosterone and DHEAS. Conclusion: Our findings support the hypothesis that there is association of shorter CAG repeats with Egyption PCOS patients suggesting inherited alteration in androgen sensitivity may contribute to PCOS.

P-91: Androgen Associated Gene Polymorphism(s) in Women with Polycystic Ovary Syndrome from South Indian Population

Background: Polycystic ovary syndrome (PCOS) is a heterogeneous endocrine disorder affecting 4-12% of reproductive women worldwide; characterized by chronic anovulation, clinical and/or biochemical hyperandrogenism, and polycystic ovaries on ultrasound scan. Ovarian androgen overproduction is the key pathophysiologic feature of PCOS. A number of genes encoding major enzymes of the androgen meta...

P-216: Common Polymorphisms of StAR Gene Are Not Associated with Polycystic Ovary Syndrome in Iranian Patients

Background: Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders, affecting 5-10% of reproductive aged women. High levels of androgen hormone can be accounted as the most important symptoms in PCOS. Androgen is the precursor of steroid biosynthesis in ovary. The transport of cholesterol to the inner mitochondrial membrane through StAR protein is also necessary for andr...

Role of the CAG repeat polymorphism of the androgen receptor gene in polycystic ovary syndrome (PCOS).

BACKGROUND Polycystic ovary syndrome (PCOS) is a frequent heterogenic disorder with a familial background. Androgenic effects, determining the clinical features of the syndrome, are mediated by the androgen receptor (AR), whose activity is modulated by a genetic polymorphism. We investigated the role of the CAG repeat polymorphism of the androgen receptor in PCOS. METHODS In the infertility u...

Androgen Receptor CAG Repeat Polymorphism and Epigenetic Influence among the South Indian Women with Polycystic Ovary Syndrome

The present study was carried out to assess the role of androgen receptor CAG repeat polymorphism and X chromosome inactivation (XCI) pattern among Indian PCOS women and controls which has not been hitherto explored and also to test the hypothesis that shorter CAG alleles would be preferentially activated in PCOS. CAG repeat polymorphism and X chromosome methylation patterns were compared betwe...

Investigation of adrenoceptor genes (β2 & β3) in women with polycystic ovary syndrome

Background: Genetic polymorphism is responsible for variations and individual differences. Polymorphism is a major factor of complex diseases with unknown etiology and cancer. Inconsistency in the symptoms of polycystic ovary syndrome (monthly disorder, hirsutism, obesity, diabetes, infertility) is one of the major pathological complications of this syndrome. The present study was conducted to ...
